What is the significance of Romeo's first love?

I suggest you break your answer down into parts.

Rosaline, who Romeo describes as: "The all-seeing sun / ne'er saw her match since first the world begun." is a plot device (to get Romeo to the Capulet Ball), a contrast to his real love Juliet, and a commentary on foolish, adolescent love. I would break down my answer into these three parts and expand with relevant references.

If you then want to go the extra mile try to think of something original to say!
